Contraindications:
OnlylimitedamountsofDMSOcanbeincluded,asithasbeenshowntodisruptPD-1:PD-L2interaction.AvoidgreenandbluedyesthatabsorblightintheAlphaScreensignalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).Thepresenceof>1%RPMI1640culturemediumleadstoasignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphaScreenassays.
