SorafenibTosylate,alsoknownasBay43-9006,isanovelbi-arylureacompoundthatinhibitscellproliferationbytargetingreceptortyrosinekinases,includingVEGFR-2andPDGFR-βandtheirassociatedsignalingcascadesoftheERKpathwayandangiogenesis.ItwasoriginallydevelopedasaRafkinaseinhibitor.IthasbeenshowntohaveactivityagainstseveralreceptortyrosinekinasesinvolvedintumOrigenesisandangiogenesis.SorafenibisnotactiveagainsterbB1,erbB2,ERK-1,MEK-1,EGFR,HER-2,IGFR-1,c-MET,c-yes,PKB,PKA,cdk1/cyclinB,PKC,orpim-1.Sorafenibhasbeenshowntoinhibitvariousnonkinasetargets,includingadenosineA3,dopamineD1,andmuscarineM3receptors,butatmuchhigher(micromolar)concentrationsthankinasetargets.Itisalsotheactiveingredientinadrugthathasbeenapprovedinatleastonecountryforuseinpatientswithadvancedrenalcellcancer.ThisversionisNOTforhumanuse.
