Instructionsforuse:

Preparea50mlconicaltubewith10mlofpre-warmedmedium(10%FBSinIMDMorLGM).Quicklythawcellsina37°Cwaterbathwithconstantandslowagitation.Itisimportanttoworkquicklyinthefollowingstepstoensurehighcellviabilityandrecovery.Cleantheoutsideofthevialwith70%ethanolandimmediatelytransfertheentirecontentofthetubeintothemedium.GentlyswirlthetubeandcentrifugethecellsUSPensionat300xgfor10minutesatroomtemperature.

CarefullyremovethesupernatantwithaPipettewithoutdisturbingthepellet.Gentlyresuspendthecellpelletin15-20mLofwarmmedium.Centrifugethecellsuspensionat300xgfor10minutesatroomtemperature.Carefullyremovethesupernatantwithapipettewithoutdisturbingthepellet.Gentlyresuspendthecellpelletinwarmmediumandmixbygentlyflickingthetube.NOTE:Upto30%ofcelllosscanbeexpectedduringwashingsteps.Cellsarenowreadyforuseindownstreamapplications.

Storecellsat-135˚Corcolder.Thawedcellsshouldbeusedimmediatelyfordownstreamapplications.Astheseareprimarycells,wedonotrecommendmaintainingthesecellsinculture.
