Background:

ThenuclearfactorofactivatorTcells(NFAT)familyoftranscriptionfactorsplaysanimportantroleinimmuneresponse.TcellactivationthroughtheTcellsynapseresultsincalciuminflux.Increasedintracellularcalciumlevelsactivatethecalcium-sensitivephosphatase,calcineurin,whichrapidlydephosphorylatestheserine-richregion(SRR)andSP-repeatsintheaminoterminiofNFATproteins.ThisresultsinaconformationalchangethatexposesanuclearlocalizationsignalpromotingNFATnuclearimport.Inthenucleus,NFATproteinscooperatewithotherproteinstobindtoDNA.

Description:

TheNFATReporter–JurkatCellLinecontainsafireflyluciferasegeneunderthecontroloftheNFATresponseelementstablyintegratedintoJurkatcells.TheNFATReportercelllinehasbeenvalidatedforresponsetothapsigargin,ionomycin,andphorbol12-myristate13-acetate(PMA).ItisusefulasacontrolcelllineforotherNFATreportercelllinesexpressingvariousimmunecheckpointreceptors.
