Background:
Lymphocyte-activationgene3(LAG3,CD223)isacellsurfaceproteinthatbelongstoimmunoglobulin(Ig)superfamily.LAG3isexpressedonactivatedTcells,naturalkillercells,Bcells,andplasmacytoiddendriticcells.ItsmainligandisMHCclassII,towhichitbindswithhigheraffinitythanCD4.Itnegativelyregulatescellularproliferation,activation,andhomeostasisofTcells,inasimilarfashiontoCTLA-4andPD-1,andhasbeenreportedtoplayaroleinTregsuppressivefunction.AnumberofLAG3antibodiesareinpreclinicaldevelopmentfortreatmentsforcancerandautoimmunedisorders.LAG3maybeabetterimmunecheckpointinhibitortargetthanCTLA-4orPD-1sinceantibodiestothesetwocheckpointsareonlyactivatingeffectorTcells,andnotinhibitingTregactivitywhereanantagoNISTLAG3antibodycanbothactivateeffectorTcells(bydownregulatingtheLAG3inhibitingsignal)andinhibitinduced(i.e.antigen-specific)Tregsuppressiveactivity.
