Assay Conditions:
80 µl containing 50 mM Tris pH 8.8, 5 mM MgCl2, 4 mM DTT, 10 µM substrate peptide, 20 µM S-adenosylmethionine, and 20-40 ng SUV39H1 for 1 hr. at room temperature. Add antibody against
methylated K9 residue of histone H3, incubate 1 hr. Then, add secondary HRP-labeled antibody and incubate 30 min. Finally, add HRP chemiluminescent substrates and read luminescence.

45 mM Tris-HCl, pH 8.0,
124 mM NaCl, 2.4 mM KCl, 3 mM DTT, 18
mM glutathione and 10% glycerol.
