ThekeytothePRMT9HomogeneousAssayKitisahighlyspecificantibodythatrecognizesmethylatedsubstrate.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequiredformethyltransferasedetection.First,asamplecontainingPRMT9enzymeisincubatedwiththesubstrate.Next,acceptorbeadsandprimaryantibodyareadded,thendonorbeads,followedbyreADIngtheAlpha-counts.
SuppliedAs:
KitcomesinaconvenientAlphaLISA®format,withprimaryantibody,PRMT9assaybuffer,andpurifiedPRMT9for384enzymereactions.
Contraindications:
GreenandbluedyesthatabsorblightintheAlphaScreen®signalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).Thepresenceof>1%RPMI1640culturemediumleadstoasignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphaScreen®assays.
