Pigmentepithelium-derivedfactor(PEDF),a50-kDasecretedglycoprotein,iswidelyexpressedthroughoutthehumanbody,withexpressiondecreasingduringhumanhepatocellularcarcinomaandbreastcancerprogression.PEDFbindstoacellmembranereceptortoexhibitmultifunctionalactivityinmanycelltypesbutitssignalingmechanismsarelargelyunknown.PEDFexertsanti-angiogenicactivitybyarrestingVEGF-orbFGF-mediatedendothelialcellmigration,inhibitingcapillarymorphogenesis,andinducingendothelialcellapoptosis.Inhumandermalmicrovascularcells,PEDFinducesFasLexpressionandsubsequentlyactivationofcaspase-8,whichinitiatesthedownstreamapoptoticcascade.Inhumanumbilicalveinendothelialcells(HUVECs),itsinductionofapoptosiswasshowntodependonp38MAPKactivityandtoinvolveactivationofcaspases-8and-9.
