TRPC3channelbelongstothetransientreceptorpotentialchannel(TRP)superfamilywhichisdividedintosevenmainsubfamilies.TRPchannelsshareasimilararchitectureofsix-transmembranedomainsandfunctionastetramers.Mostchannelsarepermeabletocalciumandmonovalentcations(Vassortetal.,2009).Specifically,TRPC3channelbelongstoacanonicalTRP(TRPC)subfamilythatisinvolvedincalciumentry.TRPC3isactivatedthroughphospholipaseC-linkedreceptors.WhenexpressedinHEK293cells,TRPC3behavesasareceptoractivatedchannelwithconstitutiveactivitythatcannotbefurtherincreasedbystoredepletion(Trebaketal.,2002).
