Description:
TheTIGIT:CD155HomogeneousAssayKitisdesignedtomeasuretheinhibitionofTIGITbindingtoCD155.TheTIGIT:CD155HomogeneousAssayKitcomesinaconvenientAlphaLISA®formatwithpurifiedbiotinylatedTIGIT,His-taggedCD155,andassaybuffertoperformatotalof384reactions.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequired.First,asamplecontainingTIGITandaninhibitorofchoiceisincubatedwiththeCD155for60minutes.Next,acceptorbeadsareadded,thendonorbeads,followedbyreADIngtheAlpha-counts.

Contraindications:
OnlylimitedamountsofDMSOcanbeincluded,asithasbeenshowntodisruptTIGIT:CD155interaction.AvoidgreenandbluedyesthatabsorblightintheAlphaScreensignalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).Thepresenceof>1%RPMI1640culturemediumleadstoasignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphaScreenassays.
