TheSMYD3HomogeneousAssayKitisdesignedtomeasureSMYD3activityforscreeningandprofilingapplications.SMYD3isahistonemethyltransferasethatpromotesdiandtrimethylationoflysine4ofhistoneH3.ItalsoexhibitsmethylationactivitytowardalysineresidueofMAPK/ERKKinaseKinase2(MAP3K2).ThekeytotheSMYD3HomogeneousAssayKitisahighlyspecificantibodythatrecognizesmethylatedsubstrate.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequiredformethyltransferasedetection.First,asamplecontainingSMYD3enzymeisincubatedwiththesubstrateforthreehours.Next,acceptorbeadsandprimaryantibodyareadded,thendonorbeads,followedbyreADIngtheAlpha-counts.

Contraindications:
GreenandbluedyesthatabsorblightintheAlphaScreen®signalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).Thepresenceof>1%RPMI1640culturemediumleadstoasignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphaScreen®assays.
