CD40,aTNFreceptorsuperfamilymember,wasinitiallyidentifiedonBlymphocytes.Antigenpresentingcells(APCs)suchasmonocytes,basophils,dendriticcells,havebeenfoundtoexpressCD40.Additionally,CD40hasbeenidentifiedinnon-immunecellssuchasendothelialcellsandepithelialcells.Awidevarietyofcarcinomacellsover-expressCD40aswell.InteractionwithCD40ligand(CD40L,CD154)onCD4+Thelperlymphocytestriggerstheexpressionofintercellularadhesionmolecule(ICAM)andotherpro-inflammatorycytokines.CD40:CD40Lsignalingsimultaneouslyincreasesactivationofantigen-specificTcells.CD40activatesNF-κB-dependentsignalinginresponsetolipopolysaccharide(LPS)foundonGramnegativebacterialpathogens.FurThermore,agoNISTicCD40monoclonalantibodieshavebeenshowntoactivateantigenpresentingcells(APC)andpromoteanti-tumorT-cellresponsesinadditiontofosteringcytotoxicmyeloidcellswiththepotentialtocontrolcancerintheabsenceofT-cellimmunity.
Description:
RecombinantHEK293celllineexpressingfulllengthhumanCD40(Tumornecrosisfactorreceptorsuperfamilymember5;TNFRSF5).Expressionisconfirmedbyreal-timeqPCRandWesternBlot.ThisNF-ĸBluciferasereporterconstructisstablyintegratedintothegenome.Thefireflyluciferasegeneiscontrolledby4copiesofNF-κBresponseelementlocatedupstreamoftheTATApromoter.FollowingactivationbyhumanCD40ligand,NF-ĸBtranscriptionfactorbindstotheDNAresponseelementstoinducetranscriptionoftheluciferasegene.

CultureConditions:
FrozenCells:PrepareT-25cultureflaskwith10mlofpre-warmedgrowthmedium.Quicklythawcellsina37°Cwaterbathwithconstantandslowagitation.Cleantheoutsideofthevialwith70%ethanolandimmediatelytransfertheentirecontenttotheculturemediawithoutHygromycinBandG418.Avoidpipettingupanddown,andgentlyrocktheflasktodistributethecells.Incubatethecellsinahumidified37°Cincubatorwith5%CO2.48hoursafterincubation,changetofreshmediumwithoutHygromycinBandG418,withoutdisturbingtheattachedcells.Continuetochangethemediumevery2-3daysuntilcellsreachdesiredconfluency.Ifslowcellgrowthoccursduringresuscitation,increaseFBSto15%forthefirstweekofculture.BeginaddingHygromycinBandG418tomediumafterthefirstpassage.Subculture:Whencellsreached90%confluency,removethemediumandGENTLYwashoncewithPBS(withoutMagnesiumorCalcium).ThesecellsarelooselyadherentanddetacheasilysodonotresUSPendthePBSdirectlyontothecellsurface.Treatcellswith2mlof0.25%trypsin/EDTAandincubatefor2-3minutesat37°C.Afterconfirmingcelldetachmentbylightmicroscopy,add10mlpre-warmedmediumandgentlyPipetteupanddowntodissociatecellclumps.Transfercellstoa15mlconicaltubeandcentrifugeat200xgfor5minutes.Removethemediumandre-suspendcellsin10mlofpre-warmedgrowthmedium.Dispense5mlofthecellsuspensionintoanewT75flaskcontainingpre-warmed15mlmedia.Incubatecellsinahumidified37°Cincubatorwith5%CO2.Freezecellsinfreezingmedium(10%DMSOinFBS)whencellsreach90%confluency.Cellshavebeendemonstratedtobestableforatleast15passages;BPSrecommendspreparingfrozenstockssocellsarenotusedbeyondpassage20.
