SIRTAssayDeveloperisanessentialreagentforsignalgenerationinsirtuinassaysperformedwithanyofBPSBioscience’sSIRTsubstrateorfluorimetricassaykits.Anappropriatefluorimetricsubstrate,comprisinganacetylatedlysinesidechain,isfirstincubatedwithasamplecontainingSIRTactivity(nuclearorcellularextract,purifiedenzyme,bead-boundimmunocomplex,etc.).Deacetylationofthesubstratesensitizesitsothat,inasecondstep,treatmentwiththeSIRTassaydeveloperproducesafluorophore.Nicotinamideisincludedasan‘inhibitorstop’forSirtuins.

A2xconcentrate.Productistobeused(undiluted)at1:1ratiowithSIRTreaction.

Instructionsforuse:
Ina50µLreaction,assaySIRTactivityusingSIRT(enzyme,lysate,etc.),substrate,SIRTassaybuffer,andtestinhibitor.Incubatemicrotiterplateat37°Cfor30minutes.Addanequalvolume(50µL)ofSIRTAssayDevelopertoeachwell.Incubatetheplateatroomtemperaturefor15minutes.Readsampleinamicrotiter-platereADIngfluorimetercapableofexcitationatawavelengthintherangeof350-380nmanddetectionofemittedlightintherangeof440-460nm.“Blank”value(noenzymenegativecontrol)issubtractedfromallothervalues.
Storage/StABIlity:
Store,longterm,onlyasthe2xconcentrate,at-70°C.Concentrateisstableforatleast3freeze-thawcycles,butcareshouldbetakenthatbothfreezingandthawingstepsaredonequickly(e.g.liquidN2ordryice/EtOHforfreezing,rapidfingerrubbingforthawing).Frozenstorageofsmalleraliquotsisrecommended.
