Description:
TheSirtuin6FluorogenicAssayKitisacompleteassaysystemdesignedtomeasureSIRT6activityforscreeningandprofilingapplications.Itcomesinaconvenient96-wellformat,withallthereagentsnecessaryfor100fluorescentSIRT6activitymeasurements.Inaddition,thekitincludespurifiedSIRT6enzymeandaSIRTinhibitor,Nicotinamide,foruseasapositiveandnegativecontrol,respectively.TheFluorogenicSIRT6AssayKitisbasedonauniquefluorogenicsubstrateanddevelopercombination.ThisassaymethodeliminatesdealingwiththerADIoactivity,extraction,andchromatographyaspectsoftraditionalassays.Usingthiskit,onlytwosimplestepsonamicrotiterplateareneededtoanalyzetheSIRT6activitylevel.First,theSirtuin6fluorometricsubstrate,containingamyristoylatedlysinesidechain,isincubatedwithpurifiedSIRT6enzyme.ThedemyristoylationsensitizesthesubstratesosubsequenttreatmentwiththeSIRTDeveloperproducesafluorophorethatcanthenbemeasuredusingafluorescencereader.
