GrowthdifferentiationFactor8(GDF-8),alsoknownasmyostatin,isasecretedproteinthatisexpressedspecificallyindevelopingandadultskeletalmuscle.Itcontrolsmyoblastproliferationandisapotentnegativeregulatorofskeletalmusclemass.GDF-8belongstothetransforminggrowthfactorβ(TGF-β)superfamily.AllTGF-βsuperfamilymembersaresynthesizedandsecretedasahomodimericprepropeptidethatiscleavedbyproproteinconvertasessuchasfurintogeneratethedimericN-terminalpropeptideandthedimericC-terminalmatureactiveprotein.TheC-terminalmatureproteincontainsthecharacteristicconservedcysteineresiduesinvolvedintheformationofthecysteineknotdomain.MouseGDF-8CDNAencodesa376aminoacidresidue(aa)preproproteinwithaputative24a.a.signalpeptide,a243a.a.propeptideanda109a.a.matureprotein.GDF-8ishighlyconservedacrossspecies.Maturehuman,mouse,rat,andcowGDF-8share100%aasequenceidentity.AmongTGF-βfamilymembers,GDF-8ismostcloselyrelatedtoGDF-11/BMP-11.Thetwoproteinsshare65%overallaasequence,withintheirmatureregions,thetwoproteinsdifferonlyby11a.a.residues.SimilarlytoTGF-β1,2,and3,theGDF-8homodimericpropeptideandmatureproteinremainednon-covalentlylinkedafterproteolyticcleavage,andisreleasedasaBIOLOGicallyinactivelatentcomplexthatdoesbinditsreceptor.Inserum,GDF-8hasalsobeenfoundtoexistinalargelatentcomplexthatalsoincludedFLGR(follistatin-relatedgene)andGASP-1(growthanddifferentiationfactor-associatedserumprotein-1)inadditiontothepropeptide.RecombinantGDF-8propeptideiscapableofassociatingwiththeactiveGDF-8withhighaffinitytoreconstitutethelatentcomplexandispotentGDF-8antagoNIST.
