Background:
L-tryptophan(L-Trp)isanessentialaminoacidnecessaryforproteinsynthesisinmammaliancellsandtheL-Trptokynurenine(Kyn)pathwayisfirmlyestablishedasakeyregulatorofinnateandadaptiveimmunity.CatabolismofL-TrptoKynmaintainsanimmunosuppressivemicroenvironmentbystarvingimmunecellsofL-TrpandreleasingdegradationproductsofL-Trpthathaveimmunosuppressivefunctions.Indoleamine2,3-dioxygenases(IDO1&IDO2),twooftheratelimitingenzymesinthispathway,areupregulatedinmanytumors,providingcancercellswithanavenueforimmuneevasion.
Description:
TheIDO1FluorogenicInhibitorScreeningAssayKitisdesignedtomeasureenzymeinhibitionofindoleamine2,3-dioxygenase1(IDO1).TheIDO1FluorogenicInhibitorScreeningAssayKitissimpletouseanddetectsfluorescenceatlongwavelengths,whichminimizespotentialerrorsduetocompoundinterference.Intheassay,theinhibitorandenzymeareaddedtoasamplecontainingL-Trpsubstrate.Aftera1hourincubationatroomtemperature,thefluorescencesolutionisaddedandincubatedat37°Cforfourhours.ActivityismeasuredbyreADIngsamplefluorescenceat&lamBDa;=510nmfollowingexcitationofthereactionproductatλ=400nm.
