HumanPDE7Bhasbeenstablyexpressedinahumanembryonickidney(HEK293)cellline.PDE7BexpressionwasconfirmedbyWesternblotting.ThefunctionofexpressedPDE7BwasvalidatedbycAMPdetectionassay.TheregulationofintracellularlevelofcAMPbyPDE7BinPDE7Bstably-expressedHEK293cellswascharacterizedbyacell-basedreporterassayusingpCRE-lucreportervector.pCRE-luccontainstheluciferasegenethatisunderthecontrolofcAMPresponseelement(CRE).ElevationofintracellularcAMPactivatescAMPresponseelementbindingprotein(CREB)tobindCREandinducestheexpressionofluciferase.WhencellstransientlytransfectedwithpCRE-lucreporterwereactivatedbyforskolin,thelevelofcAMPwasupregulatedinparentalHEK293cellsinducingtheexpressionoftheluciferasereporterwhereashPDE7B-HEK293cellsshowedreductioninthelevelofcAMPthatresultedinloweredexpressionofluciferase.
