Description:
ThePDE5A1AssayKitisdesignedforidentificationofPDE5A1inhibitorsusingfluorescencepolarization.TheassayisbasedonthebindingofafluorescentnucleotidemonophosphategeneratedbyPDE5Atothebindingagent.PDE5Acatalyzesthehydrolysisofthephosphodiesterbondindye-labeledcyclicguanosinemonophosphate.Nanoparticlebeadsselectivelybindthephosphategroupinthenucleotideproduct.Thisincreasesthesizeofthenucleotiderelativetounreactedcyclicguanosinemonophosphate.Inthepolarizationassay,dyemoleculeswithabsorptiontransitionvectorsparalleltothelinearlypolarizedexcitationlightareselectivelyexcited.Dyesattachedtotherapidly-rotatingcyclicmonophosphateswillobtainrandomorientationsandemitlightwithlowpolarization.Dyesattachedtotheslowly-rotatingnucleotide-beadcomplexeswillnothavetimetoreorientandthereforewillemithighlypolarizedlight.ThekeytothePDE5A1AssayKitisthespecificbindingagent.Usingthiskit,onlytwosimplestepsonamicrotiterplatearerequiredforPDE5A1reactions.First,thefluorescentlylabeledcGMPisincubatedwithasamplecontainingPDE5A1for1hour.Second,abindingagentisaddedtothereactionmixtoproduceachangeinfluorescentpolarizationthatcanthenbemeasuredusingafluorescencereader.
