Description:
ThePDE2AAssayKitisdesignedforidentificationofPDE2Ainhibitorsusingfluorescencepolarization.TheassayisbasedonthebindingofafluorescentnucleotidemonophosphategeneratedbyPDE2Atothebindingagent.PDE2Acatalyzesthehydrolysisofthephosphodiesterbondindye-labeledcyclicadenosinemonophosphate(cAMP).Nanoparticlebeadsselectivelybindthephosphategroupinthenucleotideproduct.ThisincreasesthesizeofthenucleotiderelativetounreactedcAMP.Sincethedegreeofpolarizationofafluorophoreisinverselyrelatedtoitsmolecularrotation,dyesattachedtotheslowly-rotatingnucleotide-beadcomplexeswillnothavetimetoreorientandthereforewillemithighlypolarizedlight.Conversely,dyesattachedtotherapidly-rotatingcyclicmonophosphateswillobtainrandomorientationsandemitlightwithlowpolarization.ThekeytothePDE2AAssayKitisthespecificbindingagent.Usingthiskit,onlytwosimplestepsonamicrotiterplatearerequiredforPDE2Areactions.First,thefluorescentlylabeledcAMPisincubatedwithasamplecontainingPDE2A1for1hour.Second,abindingagentisaddedtothereactionmixtoproduceachangeinfluorescentpolarizationthatcanthenbemeasuredusingafluorescencereader.
