Phosphodiesterases(PDEs)playanimportantroleinthedynamicregulationofcAMPandcGMPsignaling.PDE9Aisprimarilyexpressedinspleen,smallintestine,andbrain.PDE9inhibitorshavebeenstudiedastherapeuticsfortreatmentofcardiovasculardiseases,diabetes,andneurodegenerativedisorders.Phosphodiesterasescatalyzethehydrolysisofthephosphodiesterbondindye-labeledcyclicmonophosphates.Beadsselectivelybindthephosphategroupinthenucleotideproduct.Thisincreasesthesizeofthenucleotiderelativetounreactedcyclicmonophosphate.Inthepolarizationassay,dyemoleculeswithabsorptiontransitionvectorsparalleltothelinearly-polarizedexcitationlightareselectivelyexcited.Dyesattachedtotherapidly-rotatingcyclicmonophosphateswillobtainrandomorientationsandemitlightwithlowpolarization.Dyesattachedtotheslowly-rotatingnucleotide-beadcomplexeswillnothavetimetoreorientandthereforewillemithighlypolarizedlight.

ThePDE9AAssayKitisdesignedforidentificationofPDE9Ainhibitorsusingfluorescencepolarization.TheassayisbasedonthebindingofafluorescentnucleotidemonophosphategeneratedbyPDE9Atothebindingagent.ThekeytothePDE9AAssayKitisthespecificbindingagent.Usingthiskit,onlytwosimplestepsonamicrotiterplatearerequiredforPDE9Areactions.First,thefluorescentlylabeledcGMPisincubatedwithasamplecontainingPDE9Afor1hour.Second,abindingagentisaddedtothereactionmixtoproduceachangeinfluorescentpolarizationthatcanthenbemeasuredusingafluorescencereader.
