TheSETD2DirectActivityChemiluminescentAssayKitisdesignedtomeasureSETD2activityforscreeningandprofilingapplications.ThekeytotheSETD2ChemiluminescentAssayKitisahighlyspecificantibodythatrecognizesmethylatedK36residueofHistoneH3.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequiredformethyltransferasedetection.First,S-adenosylmethionineisincubatedwithasamplecontainingassaybufferandmethyltransferaseenzymeforonehour.Next,primaryantibodyisadded.Finally,theplateistreatedwithanHRP-labeledsecondaryantibodyfollowedbyadditionoftheHRPsubstratetoproducechemiluminescencethatcanthenbemeasuredusingachemiluminescencereader.

AvoidgreenandbluedyesthatabsorblightintheAlphascreen®signalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).Thepresenceof>1%RPMI1640culturemediumleadstoasignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphascreen®assays.
