ApremilastbindstothecatalyticsiteofPDE4,withvaluesofinhibitionconstantIC50andaffinityconstantKiof0.074µMand68nMrespectively,consequentlyleADIngtothedegradationofcAMP.Apremilastexhibitsanti-inflammatoryactivitiesagainstinflammatorydiseaseinanimalmodelsaswellashumanchronicinflammatorydiseases(suchaspsoriasisandpsoriaticarthritis)byblockingthesynthesisofarangeofpro-flammatorycytokinesandchemokines,includingtumornecrosisfactoralpha,interleukin23,CXCL9andCXCL10.
