Background:

CD155isatransmembraneglycoproteinreceptorbelongingtotheimmunoglobulinsuperfamily.Itishighlyexpressedondendriticcells,fibroblasts,endothelialcells,andmultipletumorcellsincludingovariancarcinoma,non-smallcelllung,glioblastoma,andcolorectalcarcinoma.IthasahighaffinityforaTcellregulatorytransmembranesurfaceproteincalledTIGIT(TcellimmunoglobulinandITIMdomains),andplaysakeyroleintumorcellinvasionandmigration.InteractionbetweenCD155andTIGITcausesimmunosuppressiveeffectsonCD4+andCD8+Tcells,whichhindersTcellproliferationandfunction.CD155alsointeractswithregulatoryreceptorsCD226(expressedonnaturalkiller(NK)cells,monocytesandCD4+Tcells)andCD96.WhileCD155-CD226engagementactivatesNKcellcytotoxicityandTcellresponse,CD155-CD96interactioninhibitsNKcellfunction.Duetotheseopposingregulatoryeffects,blockadeofCD155-TIGITandCD155-CD226interactionsareattractivetherapeuticstrategiesincancerimmunotherapyandautoimmunediseases,respectively.
