Description:
TheUTXHomogeneousAssayKitisdesignedtomeasuretheactivityofUTXforscreeningandprofilingapplications.ThekeytotheUTXHomogeneousAssayKitisahighlyspecificantibodythatrecognizesdemethylatedsubstrate.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequiredfordemethylaseactivitydetection.First,UTXenzymeisincubatedwiththebiotinylatedsubstrate.Next,acceptorbeadsandprimaryantibodyareadded,thendonorbeads,followedbyreADIngtheAlpha-counts.

GreenandbluedyesthatabsorblightintheAlphaScreen®signalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).ThepresenceofculturemediumRPMI1640at>1%leadstosignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphaScreen®assays.
