TheUTXChemiluminescentAssayKitisdesignedtomeasureUTXactivityforscreeningandprofilingapplications.UTX,alsoknownasKDM6A,isaJmjC-domainproteinthatexhibitsdemethylationactivitytowarddi-andtrimethyl-lysine27onhistoneH3(H3K27me2/3).ThekeytotheUTXChemiluminescentAssayKitisahighlyspecificantibodythatrecognizesdemethylatedsubstrate.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequiredfordetectionofdemethylaseactivity.First,UTXenzymeisincubatedwiththemethylatedH3peptideforonehour.Next,primaryantibodyisadded.Finally,theplateistreatedwithanHRP-labeledsecondaryantibodyfollowedbytheadditionoftheHRPsubstratetoproducechemiluminescencethatcanbemeasuredusingachemiluminescencereader.
