TheLSD2(KDM1B)HomogeneousAssayKit(Monomethyl)isdesignedtomeasuretheactivityoflysine-specificdemethylase(LSD2)forscreeningandprofilingapplications.LSD2,alsocalledKDM1B,isachromatin-modifyingenzymethatspecificallyremovesmethylgroupsfrommono-anddimethylatedLysofhistoneH3,therebyactingasacorepressoroftranscription.LSD2isapotentialtargetfordrugdevelopment.TheLSD2HomogeneousAssayKit(Monomethyl)comesinaconvenientAlphaLISA®format,withbiotinylated,mono-methylatedhistoneH3(1MeK4)peptidesubstrate,primaryantibodytodetectnon-methylatedproduct,LSD2assaybuffer,andpurifiedLSD2for384enzymereactions.ThekeytotheLSD2HomogeneousAssayKit(Monomethyl)isahighlyspecificantibodythatrecognizesdemethylatedsubstrate.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequiredfordemethylasedetection.First,asamplecontainingLSD2enzymeisincubatedwiththebiotinylatedsubstrate.Next,acceptorbeadsandprimaryantibodyareadded,thendonorbeads,followedbyreADIngtheAlpha-counts.
SuppliedAs:
TheLSD2HomogeneousAssayKit1comesinaconvenientAlphaLISA®format,withbiotinylated,mono-methylatedhistoneH3(1MeK4)peptidesubstrate,primaryantibodytodetectnon-methylatedproduct,LSD2assaybuffer,andpurifiedLSD2.
Contraindications:
GreenandbluedyesthatabsorblightintheAlphaScreensignalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).Thepresenceof>1%RPMI1640culturemediumleadstoasignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphaScreenassays.
