10μLofreactionmixturecontains50mMTris,pH8.8,50mMNaCl,0.01%Tween-20,0.5µMbiotinylatedpeptidesubstrate,andvariousamountofLSD2.Thereactionswereincubatedatroomtemperatureforonehourwithslowshaking.Afterthereaction,10μLofprimaryantibody(1:800dilution)andanti-mouseacceptorbeads(1:500dilution)wasaddedintothewellsandthereactionswereincubatedfor30min.Then10μLofStreptavidin-conjugateddonorbeads(1:125dilution)wasaddedintothewells.Thereactionswereincubatedforadditional30minatroomtemperaturefollowedbyAlphascreeningdetection.
