TheJMJD2CHomogeneousAssayKitisdesignedtomeasureactivityoftheJMJD2Cforscreeningandprofilingapplications,usingAlphaLISA®technology.ThekeytotheJMJD2CHomogeneousAssayKitisahighlyspecificantibodythatrecognizesdemethylatedsubstrate.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequiredformethyltransferasedetection.First,asamplecontainingJMJD2Cenzymeisincubatedwiththebiotinylatedsubstrate.Next,acceptorbeadsandprimaryantibodyareadded,thendonorbeads,followedbyreADIngtheAlpha-counts.

KitcomesinaconvenientAlphascreen®orAlphaLISA®format,witha384-wellplate,biotinylatedhistoneH3peptidesubstrate,primaryantibody,demethylaseassaybuffer,andpurifiedJMJD2Cfor384enzymereactions.
Contraindications:
AvoidgreenandbluedyesthatabsorblightintheAlphascreen®signalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).Thepresenceof>1%RPMI1640culturemediumleadstoasignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphascreen®assays.
