TheJHDM1DChemiluminescentAssayKitisdesignedtomeasureJHDM1Dactivityforscreeningandprofilingapplications.JHDM1D,alsoknownasKDM7AandKIAA1718,isaJumonjiC(JmjC)domain-containinghistonelysinedemethylasethatexhibitsdemethylationactivitytowarddimethyl-lysine9anddimethyl-lysine27onhistoneH3(H3K9me2andH3K27me2respectively).ThekeytotheJHDM1DChemiluminescentAssayKitisahighlyspecificantibodythatrecognizesdemethylatedsubstrate.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequiredfordetectionofdemethylaseactivity.First,JHDM1DenzymeisincubatedwiththemethylatedH3peptideforonehour.Next,primaryantibodyisadded.Finally,theplateistreatedwithanHRP-labeledsecondaryantibodyfollowedbytheadditionoftheHRPsubstratetoproducechemiluminescencethatcanbemeasuredusingachemiluminescencereader.
