HDACassaybufferisanessentialreagentforsignalgenerationinHDACassaysperformedwithanyofBPSBioscience"sHDACfluorimetricsubstratesorassaykits.Anappropriatefluorimetricsubstrate,comprisinganacetylatedlysinesidechain,isfirstincubatedwithasamplecontainingHDACactivity(nuclearorcellularextract,purifiedenzyme,bead-boundimmunocomplex,etc.)inHDACassaybuffer.Deacetylationofthesubstratesensitizesitsothat,inasecondstep,treatmentwiththeHDACassaydeveloperproducesafluorophore.TrichostatinAisincludedasan‘inhibitorstop’forclassIandIIHDACs.

Instructionsforuse:
Ina50µLreaction,assayHDACactivityusingHDAC(enzyme,lysate,etc.),substrate,HDACassaybuffer,andtestinhibitor.Incubatemicrotiterplateat37°Cfor30minutes.Addanequalvolume(50µL)ofHDACAssayDevelopertoeachwell.Incubatetheplateatroomtemperaturefor15minutes.Readsampleinamicrotiter-platereADIngfluorimetercapableofexcitationatawavelengthintherangeof350-380nmanddetectionofemittedlightintherangeof440-460nm.“Blank”value(noenzymenegativecontrol)issubtractedfromallothervalues.
