Background:
Thelowdensitylipoproteinreceptor(LDLRprotein)genefamilyconsistsofcellsurfaceproteinsinvolvedinreceptor-mediatedendocytosisofspecificligands.Lowdensitylipoprotein(LDL)isnormallyboundatthecellmembraneandtakenintothecellendingupinlysosomeswheretheproteinisdegradedandthecholesterolismadeavailableforrepressionofmicrosomalenzyme3-hydroxy-3-methylglutarylcoenzymeA(HMGCoA)reductase,therate-limitingstepincholesterolsynthesis.Atthesametime,areciprocalstimulationofcholesterolestersynthesistakesplace.Mutationsinthisgenecausetheautosomaldominantdisorder,familialhypercholesterolemia.IncaseofHIV-1infection,itfunctionsasareceptorforextracellularTatinneurons,mediatingitsinternalizationinuninfectedcells.PCSK9bindstotheepidermalgrowthfactor-likerepeatA(EGF-A)domainoftheLDLRprotein,inducingLDLRdegradation.
