TET2(UniProt/Swiss–ProtentryQ6N021)isamethylcytosinedioxygenasethatcatalyzestheconversionof5–methylcytosineto5–hydroxymethylcytosine(5–hmC).5–hmChasbeenrecentlydiscoveredinmammalianDNAandisabundantinPurkinjeneurons,granulecells,embryonicstemcells,andbraintissue,especiallyinareasthatareassociatedwithhighercognitivefunction.Althoughitspreciserolehasstilltobeshown,recentstudiesindicatethat5–hmCplaysimportantrolesdistinctfrom5–mC.Earlyevidencesuggeststhat5–hmCmayrepresentanewpathwaytodemethylateDNAinvolvingarepairmechanismconverting5–hmCtocytosine.MutationsinTET2havebeenassociatedwithmyeloproliferativediseasessuchasessentialthrombocythemia,polycythemiaveraandprimarymyelofibrosis.
