MeCP2(UniProt/Swiss–ProtentryP51608)isachromosomalproteinwithabundantbindingsitesinthechromatin.ItbelongstothefamilyofmethylCpGbindingproteinswhichalsocomprisesMBD1,MBD2,MBD3andMBD4.MeCP2canbindspecificallytomethylatedpromoters,therebyrepressingtranscription.ThistranscriptionalrepressionismediatedthroughinteractionwithhistonedeacetylaseandthecorepressorSIN3A.MeCP2alsoisessentialfordevelopment.MutationsinMeCP2arethecauseofseveraltypesofmentalretardationincludingRettsyndrome,aprogressiveneurologicaldisorderthatcausesmentalretardationinfemalesandmentalretardationsyndromicX–linkedtype13,andmayalsobeinvolvedinAngelmansyndromeandsusceptibilitytosometypesofautism.

AssayConditions:

ChIPresultsobtainedwiththeantibodydirectedagainstMeCP2
ChIPassayswereperformedusinghumanosteosarcoma(U2OS)cells,theantibodyagainstMeCP2(Cat.#25304)andoptimizedPCRprimersets.Shearedchromatinfrom1x10⁶cellsand5µgofantibodywereusedperChIPexperiment.IgG(1µg/IP)wasusedasanegativeIPcontrol.QuantitativePCRwasperformedwithprimersforthepromotersoftheZMYND10gene(usedasapositivecontrol)andCDC6gene(usedasanegativecontrol).Figure1showstherecovery,expressedasa%ofinput(therelativeamountofimmunoprecipitatedDNAcomparedtoinputDNAafterqPCRanalysis).

Determinationoftheantibodytiter
Todeterminethetiteroftheantibody,anELISAwasperformedusingaserialdilutionoftheantibodydirectedagainstMeCP2(Cat.#25304)andthecrudeserum.Theplateswerecoatedwiththepeptideusedforimmunizationoftherabbit.Byplottingtheabsorbanceagainsttheantibodydilution(Figure2),thetiterofthepurifiedantibodywasestimatedtobe:1:32,900.

WesternblotanalysisusingtheantibodydirectedagainstMeCP2
Nuclearextracts(40µg)fromHeLacellswereanalyzedbyWesternblotusingtheantibodyagainstMeCP2(Cat.#25304)diluted1:1,000inTBS-Tweencontaining5%skimmedmilk.Thepositionoftheproteinofinterestisindicatedontheright;theMarker(inkDa)isshownontheleft.
