PolyclonalantibodyraisedinrabbitagainsthumanJMJD2a(Jumonjidomaincontaining2a),usingaKLH–conjugatedsyntheticpeptidecontainingasequencefromthecentralpartoftheprotein.

Todeterminethetiteroftheantibody,anELISAwasperformedusingaserialdilutionoftheantibodydirectedagainsthumanJMJD2a(Cat.#25296),crudeserumandflow-through.Theplateswerecoatedwiththepeptideusedforimmunizationoftherabbit.Byplottingtheabsorbanceagainsttheantibodydilution(Figure1),thetiteroftheantibodywasestimatedtobe1:9,000.

WesternblotanalysisusingtheantibodydirectedagainstJMJD2a
NuclearextractsfromHeLacells(40µg)wereanalyzedbyWesternblotusingtheantibodyagainstJMJD2a(Cat.#25296)diluted1:500inTBS-Tweencontaining5%skimmedmilk.Thepositionoftheproteinofinterestisindicatedontheright(expectedsize121kDa);theMarker(inkDa)isshownontheleft.
