JARID2(UniProtKB/Swiss–ProtentryQ92833)isaregulatorofhistonemethyltransferasecomplexesthatplaysanessentialroleinembryonicdevelopment,includingheartandliverdevelopment,neuraltubefusionprocessandhematopoiesis.JARID2actsbyrecruitinghistonemethyltransferasecomplexes(PRC2)totheirtargetgenesandmodulatingthehistonemethyltransferaseactivity.Itdoesnothavehistonedemethylaseactivitybutregulatesactivityofvarioushistonemethyltransferasecomplexes.Inembryonicstemcells,itassociateswiththePRC2complexandinhibitstrimethylationofH3K27me3bythePRC2complex,therebyplayingakeyroleindifferentiationofembryonicstemcellsandnormaldevelopment.Incardiaccells,itisrequiredtorepressexpressionofcyclin–D1(CCND1)byactivatingH3K9methylationbytheGLP1/EHMT1andG9a/EHMT2histonemethyltransferases.
