ChIPassayswereperformedusingNIH3T3cellsand4µgoftheantibodydirectedagainstHP1α,ßandγ(Cat.#25292).QPCRwasperformedontheimmunoprecipitatedDNAwithoptimizedprimersetsfortherDNApromoterandforasubtelomericsequenceofchromosome19.Figure1showstherelativeenrichmentascomparedtoanoantibodynegativecontrolChIP.
WesternblotanalysisusingtheantibodydirectedagainstHP1α,ßandγ
WesternblotwasperformedonnuclearextractsfromHeLacells(20µg)withtheantibodyagainsthumanHP1α,ßandγ(Cat.#25292)diluted1:1,000inTBS-Tweencontaining5%skimmedmilk(Figure1).ThemolecularweightMarker(inkDa)isshownontheleft;theexpectedlocationofHP1α,HP1βandHP1γisindicatedontheright.
ImmunofluorescenceusingtheantibodydirectedagainstHP1α,ßandγ
HeLacellswerestainedwiththeantibodyagainstHP1α,ßandγ(Cat.#25292)andwithDAPI.Cellswerefixedwith4%formaldehydefor10min.andblockedwithPBS/TX-100containing5%normalgoatserumand1%BSA.ThecellswereimmunofluorescentlylabelledwiththeHP1α,ß,γantibody(left)diluted1:500inblockingsolutionfollowedbyananti-rabbitantibodyconjugatedtoAlexa488.ThemiddlepanelshowsstainingofthenucleiwithDAPI.Amergeofthetwostainingsisshownontheright.
Storeat–80°Cforupto2years.Centrifugeafterfirstthawtomaximizeproductrecovery.Aliquottoavoidrepeatedfreeze/thawcycles.Aliquotsmaybestoredat–20°Cforatleastonemonth.
