WesternblotwasperformedonnuclearextractsfromHeLacells(20μg)withtheantibodyagainsthumanHP1α(Cat.No.25291),diluted1:1,000inTBS-Tweencontaining5%skimmedmilk(Figure1).ThemolecularweightMarker(inkDa)isshownontheleft;theexpectedlocationofHP1α,HP1βandHP1γisindicatedontheright.ThewesternblotanalysisclearlydemonstratesthattheHP1αantibodyspecificallyrecognizesHP1αandnotHP1βandHP1γ.

ImmunofluorescenceusingtheantibodydirectedagainstHP1α
HeLacellswerestainedwiththeantibodyagainstHP1α(Cat.No.25291)andwithDAPI.Cellswerefixedwith4%formaldehydefor10minutesandblockedwithPBS/TX-100containing5%normalgoatserumand1%BSA.ThecellswereimmunofluorescentlylabelledwiththeHP1αantibody(left)diluted1:500inblockingsolutionfollowedbyananti-rabbitantibodyconjugatedtoAlexa488.ThemiddlepanelshowsstainingofthenucleiwithDAPI.Amergeofthetwostainingsisshownontheright.

Storeat–80°Cforupto2years.Centrifugeafterfirstthawtomaximizeproductrecovery.Aliquottoavoidrepeatedfreeze/thawcycles.Aliquotsmaybestoredat–20°Cforatleastonemonth.
