HDAC3(UniProt/Swiss–ProtentryO15379)catalysesthedeacetylationoflysineresiduesintheN–terminalpartofthecorehistones(H2A,H2B,H3andH4).AcetylationanddeacetylationofthesehighlyconservedlysineresiduesisimportantforthecontrolofgeneexpressionandHDACactivityisassociatedwithgenerepression.Histonedeacetylationisestablishedbytheformationoflargemultiproteincomplexes.HDAC3maybindtothezinc–fingertranscriptionfactorYY1,therebyregulatingtranscription.Itisalsoabletomodulatecellgrowthandapoptosisthroughtheinteractionwithp53andisthoughttobeessentialfortherepressionofthePOU1F1transcriptionfactor.

MonoclonalantibodyraisedinmouseagainsthumanHDAC3(Histonedeacetylase3),usingaKLH–conjugatedsyntheticpeptidecontainingasequencefromtheC–terminalregionoftheprotein.

ChIPresultsobtainedwiththemonoclonalantibodydirectedagainstHDAC3
ChIPassayswereperformedusinghumanHeLacells,themonoclonalantibodyagainstHDAC3(Cat.#25289)andoptimizedPCRprimersetsforqPCR.ChIPwasperformedonshearedchromatinfrom10,000cells.Atitrationoftheantibodyconsistingof1,2,5,and10µgperChIPexperimentwasanalyzed.IgG(5µg/IP)wasusedasnegativeIPcontrol.QPCRwasperformedwithprimersforthepromotersoftheactivegenesc-fosandGAPDH,andforthecodingregionofp21,aknowntargetgeneofHDAC3.Figure4showstherecovery,expressedasa%ofinput(therelativeamountofimmunoprecipitatedDNAcomparedtoinputDNAafterqPCRanalysis).

ImmunofluorescenceusingthemonoclonalantibodydirectedagainstHDAC3
HeLacellswerestainedwiththeantibodyagainstHDAC3(Cat.#25289)andwithDAPI.Cellswerefixedwith4%formaldehydefor10min.andblockedwithPBS/TX-100containing5%normalgoatserumand1%BSA.ThecellswereimmunofluorescentlylabelledwiththeHDAC3antibody(left)diluted1:500inblockingsolutionfollowedbyananti-mouseantibodyconjugatedtoAlexa594.ThemiddlepanelshowsstainingofthenucleiwithDAPI.Amergeofthetwostainingsisshownontheright.
