ChIPresultsobtainedwiththeantibodydirectedagainstH4pan
ChIPassayswereperformedusinghumanHeLacells,theantibodyagainstH4pan(Cat.#25278)andoptimizedPCRprimersetsforqPCR.ChIPwasperformedonshearedchromatinfrom10,000cells.Atitrationoftheantibodyconsistingof1,2,5,and10µgperChIPexperimentwasanalyzed.IgG(1µg/IP)wasusedasnegativeIPcontrol.QPCRwasperformedwithprimersfortheACTBpromoterandforthesecondexonoftheMBgene.Figure1showstherecovery,expressedasa%ofinput(therelativeamountofimmunoprecipitatedDNAcomparedtoinputDNAafterqPCRanalysis).

Determinationofthetiter
Todeterminethetiteroftheantibody,anELISAwasperformedusingaserialdilutionoftheantibodydirectedagainstH4pan(Cat.#25278),crudeserumandflow-throughinantigencoatedwells.Byplottingtheabsorbanceagainsttheantibodydilution(Figure2),thetiteroftheantibodywasestimatedtobe1:11,250.

CrossreactivitytestusingtheantibodydirectedagainstH4pan
ADotBlotanalysiswasperformedtotestthecrossreactivityoftheantibodyagainstH4pan(Cat.#25278)withthepeptideusedforimmunizationoftherabbitandotherpeptidescontainingunmodifiedsequencesofdifferenthistones.Onehundredto0.2pmoloftherespectivepeptideswerespottedonamembrane.Theantibodywasusedatadilutionof1:10,000.Figure3showsahighspecificityoftheantibodyforthespecificpeptide. 

Storeat–80°Cforupto2years.Centrifugeafterfirstthawtomaximizeproductrecovery.Aliquottoavoidrepeatedfreeze/thawcycles.Aliquotsmaybestoredat–20°Cforatleastonemonth.
