VDVADhasbeenshowntobeapreferredcleavagesiteforcaspase-2.Caspase2isaveryversatilecaspasewithmultiplefunctions.Itisamemberofthefamilyofcysteineproteasescalledcaspasesthatcleaveproteinsonlyatanaminoacidfollowinganasparticacidresidue.Withinthisfamily,caspase2ispartoftheIch-1subfamily.Caspase2hasasimilaraminoacidsequencetoinitiatorcaspases,includingcaspase1,caspase4,caspase5,andcaspase9.Itisproducedasazymogenwithalongpro-domain.Pro-caspase-2containstwosubunits,p19andp12.Caspase2associateswithseveralproteinsinvolvedinapoptosisviaitsCARDdomain,includingRAIDD,ARC,andDEFCAP.TogetherwithRAIDDandPIDD/LRDD,caspase2hasbeenshowntoformthePIDDosome,whichmayserveasanactivationplatformfortheprotease,althoughitmayalsobeactivatedintheabsenceofPIDD.Caspase2mayalsoformacomplexwiththeDNA-PKcsandPIDD/LRDD,whichisinvolvedinDNArepairafterDNAdamage.

Z-VDVAD-FMKisasyntheticpeptidethatirreversIBLyinhibitstheactivityofcaspase-2.Itattenuatesoxyhemoglobin-inducedcleavageofPARPandapoptosisinendothelialcells.Z-VDVAD-FMKcanbeusedforbothinvitroandinvivostudies--seenoteforusingthisproductinvitro.
