Phosphodiesterases(PDEs)playanimportantroleinthedynamicregulationofcAMPandcGMPsignaling.PDE8isinvolvedinmanyBIOLOGicalprocesses,includingT-cellactivation,testosteroneproduction,adrenocorticalhyperplasia,andthyroidfunction.Phosphodiesterasescatalyzethehydrolysisofthephosphodiesterbondindye-labeledcyclicmonophosphates.Beadsselectivelybindthephosphategroupinthenucleotideproduct.Thisincreasesthesizeofthenucleotiderelativetounreactedcyclicmonophosphate.Inthepolarizationassay,dyemoleculeswithabsorptiontransitionvectorsparalleltothelinearly-polarizedexcitationlightareselectivelyexcited.Dyesattachedtotherapidly-rotatingcyclicmonophosphateswillobtainrandomorientationsandemitlightwithlowpolarization.Dyesattachedtotheslowly-rotatingnucleotide-beadcomplexeswillnothavetimetoreorientandthereforewillemithighlypolarizedlight.

ThePDE8AAssayKitisdesignedforidentificationofPDE8Ainhibitorsusingfluorescencepolarization.TheassayisbasedonthebindingofafluorescentnucleotidemonophosphategeneratedbyPDE8Atothebindingagent.ThekeytothePDE8AAssayKitisthespecificbindingagent.Usingthiskit,onlytwosimplestepsonamicrotiterplatearerequiredforPDE8Areactions.First,thefluorescentlylabeledcAMPisincubatedwithasamplecontainingPDE8Afor1hour.Second,abindingagentisaddedtothereactionmixtoproduceachangeinfluorescentpolarizationthatcanthenbemeasuredusingafluorescencereader.
