Phosphodiesterases(PDEs)playanimportantroleinthedynamicregulationofcAMPandcGMPsignaling.PDE4Disaregulatorofairwaysmooth-musclecontractility,andhasbeenidentifiedasapotentialriskpredictorforischemicstroke.Additionally,PDE4Dhasbeenassociatedwithasthmapathophysiologyandboneformation.ThePDE4Dgeneencodesatleast9differentisoforms.Phosphodiesterasescatalyzethehydrolysisofthephosphodiesterbondindye-labeledcyclicmonophosphates.Beadsselectivelybindthephosphategroupinthenucleotideproduct.Thisincreasesthesizeofthenucleotiderelativetounreactedcyclicmonophosphate.Inthepolarizationassay,dyemoleculeswithabsorptiontransitionvectorsparalleltothelinearly-polarizedexcitationlightareselectivelyexcited.Dyesattachedtotherapidly-rotatingcyclicmonophosphateswillobtainrandomorientationsandemitlightwithlowpolarization.Dyesattachedtotheslowly-rotatingnucleotide-beadcomplexeswillnothavetimetoreorientandthereforewillemithighlypolarizedlight.

ThePDE4D3AssayKitisdesignedforidentificationofinhibitorsofPDE4D3usingfluorescencepolarization.TheassayisbasedonthebindingofafluorescentnucleotidemonophosphategeneratedbyPDE4D3tothebindingagent.ThekeytothePDE4D3AssayKitisthespecificbindingagent.Usingthiskit,onlytwosimplestepsonamicrotiterplatearerequiredforPDE4D3reactions.First,thefluorescentlylabeledcAMPisincubatedwithasamplecontainingPDE4D3for1hour.Second,abindingagentisaddedtothereactionmixtoproduceachangeinfluorescentpolarizationthatcanthenbemeasuredusingafluorescencereaderequippedforthemeasurementoffluorescencepolarization.
