Phosphodiesterases(PDEs)playanimportantroleinthedynamicregulationofcAMPandcGMPsignaling.PDE1Bisacalcium-dependentcyclicnucleotidephosphodiesterasethatishighlyexpressedinthestriatum.Itplaysthephysiologicalroleinthecentralnervoussystem.Phosphodiesterasescatalyzethehydrolysisofthephosphodiesterbondindye-labeledcyclicmonophosphates.Beadsselectivelybindthephosphategroupinthenucleotideproduct.Thisincreasesthesizeofthenucleotiderelativetounreactedcyclicmonophosphate.Inthepolarizationassay,dyemoleculeswithabsorptiontransitionvectorsparalleltothelinearly-polarizedexcitationlightareselectivelyexcited.Dyesattachedtotherapidly-rotatingcyclicmonophosphateswillobtainrandomorientationsandemitlightwithlowpolarization.Dyesattachedtotheslowly-rotatingnucleotide-beadcomplexeswillnothavetimetoreorientandthereforewillemithighlypolarizedlight.

ThePDE1BAssayKitisdesignedforidentificationofPDE1Binhibitorsusingfluorescencepolarization.TheassayisbasedonthebindingofafluorescentnucleotidemonophosphategeneratedbyPDE1Btothebindingagent.ThekeytothePDE1BAssayKitisthespecificbindingagent.Usingthiskit,onlytwosimplestepsonamicrotiterplatearerequiredforPDE1Breactions.First,thefluorescentlylabeledcAMPisincubatedwithasamplecontainingPDE1Bfor1hour.Second,abindingagentisaddedtothereactionmixtoproduceachangeinfluorescentpolarizationthatcanthenbemeasuredusingafluorescencereaderequippedforthemeasurementoffluorescencepolarization.
