Phosphodiesterases(PDEs)areinvolvedinthedynamicregulationofcAMPandcGMPsignaling.PDE11Ahasbeenobservedinskeletalmuscle,prostate,testis,salivarygland,thyroidgland,andliver.PDE11AmayplayroleintheCNSwithanassociationtomajordepressivedisorder.Phosphodiesterasescatalyzethehydrolysisofthephosphodiesterbondindye-labeledcyclicmonophosphates.Beadsselectivelybindthephosphategroupinthenucleotideproduct.Thisincreasesthesizeofthenucleotiderelativetounreactedcyclicmonophosphate.Inthepolarizationassay,dyemoleculeswithabsorptiontransitionvectorsparalleltothelinearly-polarizedexcitationlightareselectivelyexcited.Dyesattachedtotherapidly-rotatingcyclicmonophosphateswillobtainrandomorientationsandemitlightwithlowpolarization.Dyesattachedtotheslowly-rotatingnucleotide-beadcomplexeswillnothavetimetoreorientandthereforewillemithighlypolarizedlight.

ThePDE11AAssayKitisdesignedforidentificationofPDE11Ainhibitorsusingfluorescencepolarization.TheassayisbasedonthebindingofafluorescentnucleotidemonophosphategeneratedbyPDE11Atothebindingagent.ThekeytothePDE11AAssayKitisthespecificbindingagent.Usingthiskit,onlytwosimplestepsonamicrotiterplatearerequiredforPDE11Areactions.First,thefluorescentlylabeledcAMPisincubatedwithasamplecontainingPDE11Afor1hour.Second,abindingagentisaddedtothereactionmixtoproduceachangeinfluorescentpolarizationthatcanthenbemeasuredusingafluorescencereader.
