N-terminalHis-taggedhumanPDE10A2hasbeenstablyexpressedinahumanembryonickidney(HEK293)cellline.PDE10A2expressionwasconfirmedbyWesternblotting.ThefunctionofPDE10A2wasconfirmedbycAMPdetectionassay.ForskoliniscommonlyusedtoraiselevelsofintracellularcyclicAMP(cAMP).WhenHEK293cellsareactivatedbyforskolin,cAMPlevelsareupregulatedinparentalcells,whereascellsoverexpressingPDE10AshowreductionincAMPaccumulation.InhibitionofPDE10AactivityrestorescAMPlevels.
