Phosphodiesterases(PDEs)playanimportantroleinthedynamicregulationofcAMPandcGMPsignaling.PDE10AisadualsubstratePDEhighlyexpressedinstriatalmediumspinyneurons.PDE10Ainhibitorscanimprovethecognitivesymptomsofschizophrenia,andexhibitpotentialtherapeuticvalueforHuntington’sDisease.Phosphodiesterasescatalyzethehydrolysisofthephosphodiesterbondindye-labeledcyclicmonophosphates.Beadsselectivelybindthephosphategroupinthenucleotideproduct.Thisincreasesthesizeofthenucleotiderelativetounreactedcyclicmonophosphate.Inthepolarizationassay,dyemoleculeswithabsorptiontransitionvectorsparalleltothelinearly-polarizedexcitationlightareselectivelyexcited.Dyesattachedtotherapidly-rotatingcyclicmonophosphateswillobtainrandomorientationsandemitlightwithlowpolarization.Dyesattachedtotheslowly-rotatingnucleotide-beadcomplexeswillnothavetimetoreorientandthereforewillemithighlypolarizedlight.

ThePDE10AAssayKitisdesignedforidentificationofPDE10Ainhibitorsusingfluorescencepolarization.TheassayisbasedonthebindingofafluorescentnucleotidemonophosphategeneratedbyPDE10Atothebindingagent.ThekeytothePDE10AAssayKitisthespecificbindingagent.Usingthiskit,onlytwosimplestepsonamicrotiterplatearerequiredforPDE10Areactions.First,thefluorescentlylabeledcAMPisincubatedwithasamplecontainingPDE10Afor1hour.Second,abindingagentisaddedtothereactionmixtoproduceachangeinfluorescentpolarizationthatcanthenbemeasuredusingafluorescencereaderequippedforthemeasurementoffluorescencepolarization.
