TheMLL1ComplexDirectActivityChemiluminescentAssaykitisdesignedtomeasureMLL1activityforscreeningandprofilingapplications,usingpurifiedMLL1anditscomplexcomponents:WDR5,RbBP5,Ash2,andDPY30.ThekeytotheMLL1ComplexChemiluminescentAssayKitisahighlyspecificantibodythatrecognizesmethylatedK4residueofHistoneH3.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequiredformethyltransferasedetection.First,S-adenosylmethionineisincubatedwithasamplecontainingassaybufferandmethyltransferaseenzymeforonehour.Next,primaryantibodyisadded.Finally,theplatesaretreatedwithanHRP-labeledsecondaryantibodyfollowedbytheadditionoftheHRPsubstratetoproducechemiluminescencethatcanbemeasuredusingachemiluminescencereader.
