HistoneH4(R3)methyltransferasesareenzymesthatcatalyzethetransferofamethylgroupfromthecofactorS-adenosylmethioninetoarginine3residueofhistoneH4.ThisgroupofenzymesincludesPRMT1,PRMT3,andPRMT5.
Description:
TheHistoneH4(R3)UniversalMethyltransferaseAssayKitisdesignedforthedetectionofhistoneH4(R3)methyltransferase(HMT)activityinscreeningandprofilingapplications.ThekeytotheHistoneH4(R3)UniversalMethyltransferaseAssayKitisahighlyspecificantibodythatrecognizesmethylatedR3residueofHistoneH4.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequiredformethyltransferasedetection.First,S-adenosylmethionineisincubatedwithasamplecontainingassaybufferandmethyltransferaseenzymeforonehour.Next,primaryantibodyisadded.Finally,theplatesaretreatedwithanHRP-labeledsecondaryantibodyfollowedbyadditionoftheHRPsubstratetoproducechemiluminescencethatcanthenbemeasuredusingachemiluminescencereader.
SuppliedAs:
Kitcomesinaconvenientformat,witha96-wellplateprecoatedwithhistoneH4peptidesubstrate,purifiedPRMT1,PRMT3andPRMT5enzymes,andreagentsfor100enzymereactions.
Contraindications:
AvoidgreenandbluedyesthatabsorblightintheAlphascreen®signalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).Thepresenceof>1%RPMI1640culturemediumleadstoasignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphascreen®assays.
