TheG9aHomogeneousAssayKitisdesignedtomeasuretheactivityofG9a(alsoknownasEHMT2orKMT1C)forscreeningandprofilingapplications,usingAlphaLISA®technology.ThekeytotheG9aHomogeneousAssayKitisahighlyspecificantibodythatrecognizesmethylatedsubstrate.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequiredformethyltransferasedetection.First,asamplecontainingG9aenzymeisincubatedwiththebiotinylatedsubstrateforonehour.Next,acceptorbeadsandprimaryantibodyareadded,thendonorbeads,followedbyreADIngtheAlpha-counts.

SuppliedAs:

Contraindications:
AvoidgreenandbluedyesthatabsorblightintheAlphascreen®signalemissionrange(520-620nm),suchasTrypanBlue.Avoidtheuseofthepotentsingletoxygenquencherssuchassodiumazide(NaN3)ormetalions(Fe2+,Fe3+,Cu2+,Zn2+andNi2+).Thepresenceof>1%RPMI1640culturemediumleadstoasignalreductionduetothepresenceofexcessbiotinandironinthismedium.MEM,whichlacksthesecomponents,doesnotaffectAlphascreen®assays.
