TheEZH2(Y641N)TR-FRETAssayKitisdesignedtomeasureactivityofthemutantEZH2complex[EZH2(Y641N)/EED/SUZ12/RbAp48/AEBP]inahomogeneous384reactionformat.ThisFRET-basedassayrequiresnotime-consumingwashingsteps,makingitespeciallysuitableforhighthroughputscreeningapplications.ThekeytotheEZH2(Y641N)AssayKitisahighlyspecificantibodythatrecognizesmethylatedHistoneH3K27.Withthiskit,onlythreesimplestepsonamicrotiterplatearerequiredformethyltransferasedetection.First,Sadenosylmethionineisincubatedwithasamplecontainingassaybufferandmethyltransferaseenzymefor4hours.Next,antibodytomethylatedH3K27isadded.Finally,dye-labeledacceptorisaddedfollowedbyfluorescencedetection.
